What is an Adverse Outcome Pathway (AOP)?
A conceptual framework that portrays existing knowledge concerning the key events (KE) that causally link a molecular initiating event (MIE) to an adverse outcome (AO).The five AOP principles
- AOPs are not chemical-specific
- AOPs are modular and composed of reusable components
- An AOP is a pragmatic unit of development and evaluation
- For most real‐world applications, AOP networks are the functional units of prediction
- AOPs are living documents
AOPs can be used for, e.g:
- Structuring current knowledge and integrating different types of data
- Identifying data gaps
- Predicting toxicity based on mechanistic knowledge
- Strategic planning and testing, IATA
- Support development of new toxicity test methods.
- Investigating how chemicals in a mixture may interact
